Comparsion of two labeling methods showed that 99mTc-recombinant hirudin (99mTc-rH) prepared by modified 2-imiothiolane method had higher radiochemical purity and stability than that by direct labeling method. Biodistribution of 99mTc-rH in mice showed that the radioactivity in heart
lung
liver and spleen was very low
while that in kidney was high. 99mTc-rH was cleared rapidly in blood and excreted mostly from kidney. The arterial and venous thrombi in rabbits were clearly imaged 60min after injection of 99mTc-rH
T/B ratio was 2.24 and 1.95 respectively. The rH would be a potential agent for diagnosis of thrombotic disease.
